| Abstract: | Novos contributos para o estudo da eficiência da embriogénese somática em
Olea europaea L. - Resumo: embriogénese somática (ES) apresenta-se como uma estratégia alternativa para a propagação clonal
de oliveira, com particular interesse em genótipos caracterizados por elevada recalcitrância ao
desenvolvimento de raízes adventícias quando propagados pelos métodos convencionais. No entanto, a
elevada recalcitrância dos tecidos adultos à indução de estruturas embriogénicas continua a restringir a
aplicação desta tecnologia. Assim, o estudo dos mecanismos moleculares associados à resposta
embriogénica assume particular relevância para o desenvolvimento de sistemas de propagação
alternativos aplicáveis à olivicultura nacional. A ES constitui uma resposta morfogénica induzida por
estímulos exógenos que envolve uma reprogramação celular ao nível molecular, refletida em alterações
bioquímicas e fisiológicas que culminam na diferenciação de estruturas embriogénicas. Neste processo,
a interação entre os explantes e o meio de cultura é determinante, incluindo o contacto com biomoléculas
secretadas pelo material vegetal. Este intercâmbio de moléculas, nomeadamente proteínas e metabolitos,
entre os espaços intra e extracelulares poderá participar na modulação da resposta embriogénica. O
presente estudo teve como objetivo identificar mecanismos envolvidos na eficiência da ES em oliveira
através da caracterização do proteoma de linhas embriogénicas de alta (HE) e baixa (LE) eficiência e
correspondente secretoma, por LC-MS/MS, complementada pela análise do exometaboloma por 1H
RMN. A capacidade das biomoléculas secretadas para induzir a resposta embriogénica foi avaliada
através da aplicação de meio de cultura (CM) de linhas HE a linhas LE e material adulto. Os resultados
revelaram uma maior abundância de proteínas associadas ao transporte vesicular, sinalização e
reorganização celular em linhas HE, que apresentaram, no perfil extracelular, proteínas relacionadas
com a regulação redox e a dinâmica da parede celular, bem como uma utilização mais eficiente do
carbono. A aplicação do CM permitiu validar o papel das moléculas secretadas na resposta embriogénica
ao promover um aumento da proliferação celular e a formação de estruturas embriogénicas em material
recalcitrante; Somatic embryogenesis (SE) has emerged as an alternative strategy for the clonal propagation of olive
trees, particularly for genotypes characterised by high recalcitrance to adventitious root development
when propagated using conventional methods. However, the high recalcitrance of adult tissues to the
induction of embryogenic structures continues to limit the application of this technology. Therefore,
understanding the molecular mechanisms associated with the embryogenic response is of particular
importance for the development of alternative propagation systems applicable to the national olive
industry. SE is a morphogenic response induced by exogenous stimuli that involves cellular
reprogramming at the molecular level, reflected in biochemical and physiological changes that
culminate in the differentiation of embryogenic structures. During this process, the interaction between
explants and the culture medium is crucial, including exposure to biomolecules secreted by plant tissues.
This exchange of molecules, particularly proteins and metabolites, between intracellular and
extracellular compartments may contribute to the modulation of the embryogenic response. The present
study aimed to identify mechanisms involved in SE efficiency in olive through the characterisation of
the proteome of high-efficiency (HE) and low-efficiency (LE) embryogenic lines and their
corresponding secretome by LC–MS/MS, complemented by exometabolome analysis using ¹H NMR.
The ability of secreted biomolecules to induce the embryogenic response was assessed by applying
conditioned medium (CM) from HE lines to LE lines and adult material. The results revealed a higher
abundance of proteins associated with vesicular transport, signalling, and cellular reorganisation in HE
lines, which also exhibited, at the extracellular level, proteins related to redox regulation and cell wall
dynamics, together with more efficient carbon utilisation. Application of the CM validated the role of
secreted molecules in the embryogenic response by promoting increased cell proliferation and the
formation of embryogenic structures in recalcitrant material. |
